The objective of this work was to study the effect of arginine vasopressin (AVP) on contraction of isolated rat miometrium in a superfused system. For this purpose, female Sprague-Dawley rats in different stages of therir sex cycle were killed and the uterus cut into 1 cm strips. Strips were placed in Krebs-Ringer bicarbonate buffer at 37°C with continuous carbogen bubbling. Changes in contractil tonic tension (CTT) were measured with a transductor coupled to an inscriptor oscilograph. Precharge was 500 mg. The results demonstrated a significant stimulation of miometrial constraction (p < 0.05) with AVP (1.7 Units) in proestrous (900 ± 150) and estrous (900 ± 150) rats with respect to diestrous (300 10) or ovariectomized animals (112 ± 10). Progesterone added in vitro at the concentration of (0,2 ng/ml) induced a significant reduction in CTT of AVP stimulated miometrial contraction (560). Conclusions: It is known that gonadal steroids modulate AVP release from the central nervous system and pituitary. The present communication suggests that peripheral effects of AVP on contraction of rat uterus in vitro is also subject to modulation by gonadal steroids, according to the variations registered in CTT during the sex cycle and the progesterone effect.
