Abstract:

The late pathway of aldosterone production (conversion of 11-deoxycorticosterone into aldosterone) in mitochondria of rat adrenal zona glomerulosa is catalyzed by CYP11B2. In zona fasciculate, CYP11B1 transforms 11-deoxycorticosterone into corticosterone or 18-hydroxy-11-deoxycorticosterone. Homology between CYP11B1 and CYP11B2 is very high, both genes being located in tandem in chromosome 8q22. Mutations in the CYP11B2 gene and genetic recombination between CYP11B1 and CYP11B2 could be responsible for alterations in the enzymology of aldosterone production, raising a new nomenclature and explanations for the deficiencies primarily described, as CMOI and CMOII.
