Increased dopamine (DA) and noradrenaline (NA) metabolites in whole brain as well as increased DA release in slices of caudate nucleus from ethanol dependent rats have been previously found. It is known that both DA and NA are present in caudate (CAU) and accumbens (ACC) nuclei. In view of this, possible relationships were explored between chronic ethanol administration, DA and NA metabolism in CAU and ACC nuclei and despair behaviour (depression). A liquid diet with or without ethanol was given to rats for 20 consecutive days. The turnovers of DA and NA were determined by an enzymatic-isotopic micromethod after alpha-methyl-p-tyrosine. Despair behaviour was measured as the immobility of rats in an animal model for depression, swimming test (ST). Chronic ethanol enhanced DA and NA turnovers in ACC and CAU nuclei in contrast to findings in the acute ethanol groups. Chronic ethanol decreased the duration of immobility in the ST apparently through the activation of alpha1-adrenoreceptors. This effect is possibly related to an antidepressant influence of chronic ethanol which requires further investigation.
