Abstract:

Cyanobacteria are prokaryotic photosynthetic micro-organisms that produce a wide array of bioactive substances, including plant growth regulators. Many substances have been added to plant tissue culture media in order to promote plant regeneration. We studied the effect of extracellular products from Microchaete tenera Thuret ex. Born. et Flah. strain 14b on regeneration, pigments content and histology of rice callus. Calli obtained from mature embryos of Oryza sativa L. cv. Yerua PA in Murashige & Skoog's medium + 3mg/l 2,4-dichlorophenoxyacetic acid were cultivated in regeneration media, Murashige & Skoog's medium + extracellular products or Murashige & Skoog's medium + 0.5mg/l 6-benzyladenine. Histological studies confirmed the presence of shoots and roots in calli treated with cyanobacterial extracellular products and only roots with 6-benzyladenine. Chlorophyll a, b and total, and carotenoids contents in extracellular products-treated calli were: 5.01 ± 0.41, 1.81 ± 0.20, 7.87 ± 0.65 and 1.97 ± 0.21 (μg/g fresh weight), respectively and chlorophyll a/b and carotenoids/total chlorophyll ratio, 2.77 ± 0.19 and 0.25 ± 0.02 respectively. With 6-benzyladenine the values were 14.21 ± 1.40, 17.21 ± 1.68, 31.33 ± 0.28 and 3.92 ± 0.28 and 0.82 ± 0.10 and 0.12 ± 0.01, respectively. Biochemical studies indicate that chlorophyll content is not positively correlated with organogenesis. Organogenesis induction is consistent with the probable presence of plant growth regulators in cyanobacterial extracellular products which could result in beneficial effects when applied to agricultural practice.
