Abstract:

Dose-response curves of the positive inotropic effect of PGF2α on isolated rat atria, were examined. It was found a distinct efficacy stimulating the contractile tension of isolated rat atria. The compound elicited a positive inotropism which was augmented in the presence of (-)-propranolol, subthreshold doses of methoxamine, as well as after a treatment with 6-OHDA. On the contrary subthreshold concentrations of norepinephrine or the presence of phentolamine decreased the augmenting inotropic effect of PGF2α. The results suggest that the action of PGF2α is apparently direct and not mediated by the release of the tissue catecholamines. However, adrenergic mechanism appear to be somehow involved in the inotropic influence of PGF2α on the rat atria. © 1981 The Italian Pharmacological Society.
