Resumen:
Los polimorfonucleares neutrófilos (PMN) son la primera línea celular de defensa antibacteriana del organismo. Son capaces de detectar patógenos a través del reconocimiento de patrones moleculares asociados a patógenos (PAMPs, pathogen-associated molecular patterns) por medio de receptores tales como los receptores de tipo Toll (TLR, Toll-like receptors). Algunos de estos TLRs y otros receptores reconocen moléculas asociadas a la viabilidad bacteriana (denominadas vita-PAMPs). En este sentido, el ARN mensajero procariota ha sido reportado como un vita-PAMP. El objetivo de este estudio fue investigar si los PMN son capaces de censar la viabilidad bacteriana y evaluar si la presencia de ARN procariota es necesaria y suficiente para inducir una activación de los PMN y una consiguiente respuesta bactericida. Para ello, se evaluaron diferentes parámetros funcionales en PMN humanos purificados usando Escherichia coli viva (Ec) y Ec muerta por calor (MC-Ec), observándose que la viabilidad bacteriana es indispensable para inducir la quimiotaxis, la activación (aumento de tamaño y de la molécula de superficie CD11b) y las funciones bactericidas (estallido respiratorio y formación de trampas extracelulares de PMN o NETosis) en PMN. Por otro lado, el ARN procariota total purificado de Ec (ARNp) provocó respuestas similares a las observadas con las bacterias vivas. Estos hallazgos se relacionaron a la ausencia de ARNp dentro de la bacteria muerta y su liberación al sobrenadante durante el proceso de muerte, explicando la falta de respuesta de los PMN frente a las MC-Ec. Dada la relevancia que existe entre la interacción de los PMN y el endotelio en la resolución de infecciones, cuando evaluamos la capacidad del ARNp de activar directamente células endoteliales encontramos un aumento en la expresión de la molécula CD54 (ICAM-1) y una importante secreción de IL-8. Estos efectos fueron asociados con un aumento en la quimiotaxis, adhesión y transmigración de los PMN en respuesta al endotelio tratado. En conclusión, nuestros resultados indican que tanto el PMN como el endotelio son capaces de reconocer ARNp, una molécula asociada a la viabilidad bacteriana. Este reconocimiento dispara mecanismos de activación en ambos tipos celulares que modulan su inter-relación favoreciendo la adhesión de los PMN al endotelio, su transmigración, y gatillando luego mecanismos bactericidas que, en un contexto fisiológico, contribuirían a la resolución de un foco infeccioso.
Abstract:
Polymorphonuclear neutrophils (PMN) are the first cellular line of anti-bacterial host defense. They sense pathogens through recognition of pathogen-associated molecular patterns (PAMPs) by receptors, such as Toll-like receptors (TLRs). Some of these TLRs and other receptors recognize molecules associated with bacterial viability (called vita-PAMPs). Moreover, prokaryotic messenger RNA has been characterized as a vita-PAMP. Thus, the objective of this study was to investigate whether PMNs are capable of sensing bacterial viability and assess whether the presence of prokaryotic RNA is necessary and sufficient to induce PMN activation and a consequent bactericidal response. For this purpose, different functional parameters in isolated human PMN were evaluated using live Escherichia coli (Ec) and heat killed Ec (HK-Ec). We found that bacterial viability is indispensable to induce chemotaxis, activation (increase in cell size and CD11b up-regulation) and bactericidal functions (respiratory burst and neutrophil extracellular traps formation or NETosis) in PMN. Additionally, when total prokaryotic RNA of Ec (pRNA) was purified and used as a stimulus for PMN, we observed that pRNA caused similar responses to those observed with live bacteria. These findings were related to the absence of pRNA within dead bacteria and its release to the supernatant during the dying process, explaining the lack of response of PMN towards HK-Ec. Moreover, given the relevance that exists between the interaction of PMN and the endothelium in the resolution of infections, we also evaluated whether pRNA was able to directly activate endothelial cells, and whether this results in the modulation of PMN-endothelial cell interaction. We found that pRNA was able to increase the expression of CD54 (ICAM-1) and induced an important IL-8 secretion from endothelial cells. These effects were associated with an increase in chemotaxis, adhesion and transmigration of PMN in response to pRNA treated-endothelium. In conclusion, our results indicate that both PMN and endothelial cells are able to recognize pRNA, a molecule associated with bacterial viability. This recognition triggers mechanisms of activation in both cell types that modulate their inter-relation favoring PMN-endothelium adhesion and transmigration, and triggering bactericidal mechanisms in PMN that, in a physiological context, would contribute to the resolution of an infectious focus.
