Resumen:
Las mucinas de T. cruzi son glicoproteínas que cumplen un importante rol en elproceso de infección. Los oligosacáridos unidos O-glicosídicamente a proteína sonaceptores de ácido siálico que no es biosintetizado por el parásito. Las unidades β-DGalpterminales son los sitios de sialilación por la trans-sialidasa del parásito. En nuestrolaboratorio hemos abordado la síntesis de oligosacáridos de la familia de mucinas quecontienen Galf. En este trabajo de Tesis se sintetizó, por primera vez, el hexasacárido βD-Galp-(1→2)-[β-D-Galp-(1→3)]-β-D-Galp-(1→6)-[β-D-Galf-(1→2)-β-D-Galf-(1→4)]-α-DGlcNAc (I) por desarrollo de una nueva estrategia de síntesis que fue aplicadaposteriormente a la síntesis del hexasacárido β-D-Galp-(1→2)-[β-D-Galp-(1→3)]-β-DGalp-(1→6)-[β-D-Galp-(1→2)-β-D-Galf-(1→4)]-α-D-GlcNAc (II). Estos oligosacáridos sonlas estructuras más grandes liberadas como alditoles por β eliminación reductivaprovenientes de mucinas de algunas cepas de T. cruzi. El hexasacárido I fue sintetizado por una estrategia convergente [3+3] basada enuna glicosidación asistida por nitrilio, utilizando el método de tricloroacetimidato. Elsintón β-D-Galf-(1→2)-β-D-Galf-(1→4)-D-GlcNAc se construyó secuencialmente desdeel extremo reductor al no reductor, utilizando bencil α-D-galactofuranósido como materialde partida de la unidad Galf interna. La elección de este nuevo precursor, obtenido enun paso de reacción a partir de la galactosa, permitió la introducción ortogonal del grupoparticipante levulinoílo en O-2. Por consiguiente, se logró la construccióndiastereoselectiva del enlace β-D-Galf-(1→4)-D-GlcNAc por el método detricloroacetimidato. Empleando una estrategia similar convergente [3+3], se sintetizó el hexasacárido II que contiene una unidad de D-Galp terminal en lugar de D-Galf sustituyendo laposición O-2 en la Galf interna. Sin embargo, la construcción del enlace β-D-Galp- (1→2)-D-Galf fue muy dificultosa y requirió un estudio exhaustivo de glicosidación condiversos donores. Únicamente por empleo del donor superarmado electrónicamente ptolil 3,4,6-tri-O-bencil-2-O-benzoíl-1-tio-β-D-galactopiranósido se logró el producto deglicosidación dando el trisacárido derivado β-D-Galp-(1→2)-β-D-Galf-(1→4)-D-GlcNAc. La síntesis química del alditol del hexasacárido I permitió confirmar la estructuradel alditol aislado del producto natural. Los oligosacáridos sintéticos I y II serán deutilidad para los estudios biosintéticos de estas estructuras cuya biosíntesis es aúndesconocida.
Abstract:
Trypanosoma cruzi mucins are glycoproteins which play an important role in theinfection process. The oligosaccharides linked O-glycosidically to protein are acceptorsof sialic acid which is not biosynthesized by the parasite. The terminal β-D-Galp units aresites of sialylation by the parasite trans-sialidase. In our laboratory, we have undertakenthe synthesis of the oligosaccharides of the mucins family containing D-Galf. In this Thesis, the hexasaccharide β-D-Galp-(1→2)-[β-D-Galp-(1→3)]-β-D-Galp-(1→6)-β-DGalf-(1→2)-β-D-Galf-(1→4)]-α-D-GlcNAc (I) was synthesized for the first time by thedevelopment of a new strategy which was further applied for the synthesis of thehexasaccharide β-D-Galp-(1→2)-[β-D-Galp-(1→3)]-β-D-Galp-(1→6)-[β-D-Galp-(1→2)-βD-Galf-(1→4)]-α-D-GlcNAc (II). These oligosaccharides are the largest structuresreleased as alditols by reductive β-elimination from mucins of some strains of T. cruzi. Hexasaccharide I was synthesized by a [3+3]-convergent strategy based on anitrile assisted glycosylation, using the trichloroacetimidate method. The synthon β-DGalf-(1→2)-β-D-Galf-(1→4)-D-GlcNAcwas sequentially constructed from the reducingend to the non-reducing end employing benzyl α-D-galactofuranoside as starting materialfor the internal Galf unit. The choice of this novel precursor, obtained in one-reaction stepfrom galactose, allowed the introduction of an orthogonal and participating levulinoylgroup at O-2. Thus, the diastereoselective construction of the β-D-Galf-(1→4)-D-GlcNAclinkage was achieved by the trichloroacetimidate method of glycosylation. Employing a similar [3+3]-convergent strategy, the hexasaccharide II containinga terminal β-D-Galp in place of a β-D-Galf unit in the O-2 position of the internal Galf unitwas synthesized. However, the construction of the β-D-Galp-(1→2)-D-Galf linkage wasvery difficult and it required an exhaustive glycosylation study with diverse donors. Onlythrough the employment of the superarmed donor p-tolyl 2-O-benzoyl-3,4,6-tri-O-benzyl1-tio-β-D-galactopyranoside,the glycosylation product was achieved giving thetrisaccharide derivative of β-D-Galp-(1→2)-β-D-Galf-(1→4)-D-GlcNAc. The chemical synthesis of hexasaccharide alditol I allowed the confirmation ofthe structure of the alditol isolated from the natural source. Synthetic oligosaccharides Iand II will be useful for biosynthetic studies of these structures whose biosynthesisremains unknown.
