Resumen:
La enzima 6,7-dimetil-8-ribitillumazina sintetasa (lumazina sintetasa, LS) cataliza el penúltimo paso en la biosíntesis de riboflavina (vitamina B2) en plantas, hongos y bacterias. Esta proteína se caracteriza por presentar una estructura cuaternaria notoriamente divergente en función de la especie estudiada, pudiendo existir en forma de pentámeros libres, decámeros (dímeros de pentámeros) o dodecámeros de pentámeros con simetría icosaédrica. El patógeno Brucella spp., una α-proteobacteria perteneciente al orden Rhizobiales, expresa dos LSs denominadas RibH1 (pentamérica) y RibH2 (decamérica), las cuales presentan una actividad catalítica muy baja en comparación con el resto de las LSs conocidas a la fecha. La enzima RibH2, en estudio desde la década pasada, despertó un gran interés por sus propiedades inmunogénicas y por su utilidad como marcador serológico en la enfermedad brucelosis. Por otro lado, RibH1 está comenzando a ser investigada ya que fue descubierta recientemente luego de la secuenciación y anotación del genoma de diversas especies de Brucella. Uno de los aportes originales más importantes de esta Tesis consistió en la resolución y el análisis de la estructura cristalográfica de ambas isoenzimas unidas a un compuesto análogo a uno de los sustratos de la reacción catalizada por las mismas. Los resultados obtenidos permitieron plantear un modelo para intentar explicar las actividades catalíticas bajas de RibH1 y RibH2 desde un punto de vista netamente estructural. Por otra parte, se llevó a cabo también un estudio de la variabilidad de la estructura cristalográfica de RibH2 en función del pH y el modelado de diversos ligandos en su sitio activo. Las grandes diferencias observadas entre RibH2 y el resto de las LSs documentadas llevó a la hipótesis de que esta proteína podría estar cumpliendo una función adicional y aún desconocida en Brucella. Para intentar dilucidar esta cuestión, los resultados obtenidos para este patógeno se complementaron con el análisis estructural y catalítico de las isoenzimas homólogas RibH1 y RibH2 codificadas por el simbionte de leguminosas Mesorhizobium loti, otro miembro del orden Rhizobiales relacionado filogenéticamente con Brucella.
Abstract:
The enzyme 6,7-dimethyl-8-ribityllumazine synthase (lumazine synthase, LS) catalyzes the penultimate step in the biosynthesis of riboflavin (vitamin B2) in plants, fungi and bacteria. The quaternary structure of this protein varies between species, existing as free pentamers, decamers (dimers of pentamers) or dodecamers of pentamers bearing icosahedral symmetry. The pathogen Brucella spp., an α-proteobacterium that belongs to the order Rhizobiales, expresses two LSs named RibH1 (pentameric) and RibH2 (decameric). Both show very low catalytic activity in comparison with other known LSs. The enzyme RibH2, which has already been studied for over 10 years, has attracted great interest due to its immunogenic properties and its value as serological marker in the disease brucellosis. On the other hand, research on RibH1 has just begun because its presence has been recently discovered due to the sequencing and annotation of genomes from some Brucella members. One of the key findings in this Thesis was the resolution and analysis of the crystallographic structures of both isoenzymes bound to a substrate analogue. These results allowed us to propose a model that may explain, from a structural point of view, the low catalytic activities observed in RibH1 and RibH2. Next, we performed studies on the variability of the crystallographic structure of RibH2 in terms of pH, and also modeled the binding of some related ligands in its active site. We found major differences between RibH2 and the rest of the LSs documented. This led us to the hypothesis that this protein may harbor an additional, yet-unknown function in Brucella. To shed light on this topic, we complemented these results with structural and catalytic studies on the homolog RibH1 and RibH2 isoenzymes codified by the legume symbiont Mesorhizobium loti, which is another member of the order Rhizobiales and is phylogenetically related to Brucella.
