Se estudió la alteración en la carga superficial de los glóbulos rojos humanos (GR) debido a la acción de la tripsina en diferentes concentraciones, lo que induce una modificación en la agregación eritrocitaria. Se analizó la cinética de agregación y desagregación de los GR utilizando el Eritroagregámetro, que se basa en la técnica de retrodifusión láser, donde la señal registrada es inversamente proporcional al tamaño de los agregados celulares. Los resultados de estas determinaciones fueron relacionados con los valores de carga aniónica eritrocitaria (CAE) obtenidos por la técnica espectrofotométrica. Además, se analizaron las posibles alteraciones de los parámetros viscoelásticos de los GR utilizando el Eritrodefórmetro, en régimen estacionario (índice de deformabilidad, viscosidad superficial de membrana y módulo elástico) y en régimen dinámico (pérdida dinámica, módulo de almacenamiento y viscocidad compleja). Se realizó el respectivo análisis estadístico de los resultados para evaluar las diferencias significativas con respecto al grupo control. Los resultados obtenidos contribuyen a la comprensión de las alteraciones reológicas encontradas en la hipertensión arterial mediante la modelización in vitro de la disminución de la carga superficial por el tratamiento con tripsina
The alteration in the surface charge of human red blood cells (RBC) due to the action of trypsin in different concentrations, which induces a change in erythrocyte aggregation, was studied. The kinetics of RBC aggregation and disaggregation were analyzed using the Erythroaggregameter, which is based on the laser backscatter technique, where the signal recorded is inversely proportional to the size of the cell aggregates. The results of these determinations were related to the erythrocyte anionic charge (ERC) values obtained by the spectrophotometric technique. In addition, the possible alterations of the viscoelastic parameters of the RBCs were analyzed using the Erythrodephormeter, in a steady state (deformability index, superficial membrane viscosity and elastic modulus) and in a dynamic regime (dynamic loss, storage modulus and complex viscosity). The respective statistical analysis of the results was carried out to evaluate the significant differences with respect to the control group. The results obtained contribute to the understanding of the rheological alterations found in arterial hypertension through in vitro modeling of the decrease in surface charge due to trypsin treatment
