Resumen:
A la fecha, se conocen 24 variantes de la Hormona Liberadora de Gonadotrofinas (GnRH). Se ha demostrado en numerosos vertebrados,incluyendo mamíferos, que más de una variante de GnRH son expresadas en una misma especie. En la rata existen evidencias postulando la presencia de diferentes variantes de GnRH, conjuntamente con la ya caracterizada GnRH-I, ellas son: GnRH-II, gpGnRH, sGnRH y lGnRH-III. Por otro lado, se demostró la presencia de más de un receptor de GnRH (GnRHR) en distintas especies. Sin embargo, en un modelo ampliamente utilizado como lo es la rata, hay controversia acerca de los sistemas de GnRH presentes. El objetivo de esta Tesis, consiste en estudiar los distintos sistemas de GnRH presentes en rata. En este trabajo estudiamos extractos peptídicos de distintos tejidos mediante diferentes sistemas de RP-HPLC conjugado con detección con distintos RIA para GnRH, determinamos la actividad biológica de las diferentes variantes postuladas en rata y abordamos la búsqueda de un receptor diferente del conocido GnRHR tipo-I. Concluimos que, la actividad gonadotrófica hipofisaria parece estar mediada por el sistema GnRH-I/GnRHR tipo I. Demostramos la presencia de HypGnRH, aportamos fuertes evidencias de la expresión de GnRH-II en cerebro e hipófisis y postulamos la presencia de gpGnRH. No encontramos evidencias de la expresión de lGnRH-III o sGnRH. Establecimos que GnRH-I y GnRH-II poseen similares potencias en cuanto a su actividad hipofisaria, mientras que sGnRH y gpGnRH poseen una menor potencia. No detectamos efectos fisiológicos de lGnRH-III en células hipofisarias. Proponemos la existencia en células ováricas de un sitio de unión a GnRH diferente del expresado en hipófisis. Este trabajo evidencia la existencia de distintos sistemas de GnRH presentes en rata. Estos sistemas podrían participar en diferentes funciones como ser la modulación de la conducta sexual (ej GnRH-II), la regulación autócrina/parácrina de la respuesta gonadotrófica hipofisaria, regulación intra-ovárica de la función gonadal.
Abstract:
To date, 24 Gonadotropin Releasing Hormone (GnRH) variants have been described. In several vertebrates, including mammals, more than one GnRH variant are co-expressed in a single species. In the rat, there are evidences postulating various GnRH variants, together with GnRH-I, they are: GnRH-II, gpGnRH, sGnRH and lGnRH-III. On the other hand, different receptors for this hormone have been demonstrated in several species. However, in such an important model as the rat, there is controversy regarding the GnRH systems present. The aim of this Thesis was to study the different GnRH systems in the rat. Here, we characterize peptidic extracts from rat tissues by different RP-HPLC systems together with GnRH RIA detection; we study the biologic activity of the GnRH variants postulated to be present in rat and examine the possible existence of a receptor distinct from the already known type-I GnRHR. We conclude that gonadotropic activity in the rat is mediated by the GnRH-I/type-I GnRHR system. In addition, we demonstrate the expression of hypGnRH, provide strong evidences for GnRH-II presence in brain and pituitary and show experimental data supporting gpGnRH existence in the rat. However, we do not detect either lGnRH-III or sGnRH. GnRH-I and GnRH-II show similar potencies regarding gonadotropins secretion, intracellular IP3 production and Ca2+ mobilization, while sGnRH and gpGnRH are less potent. We do not detect any lGnRH-III physiologic response in rat pituitary cells. Furthermore, we propose a GnRHR different from the type-I receptor in rat ovarian SPO cells. This work demonstrates the existence of different GnRH systems in the rat, which could participate in the modulation of sexual behavior (eg. GnRH-II), autocrine/paracrine regulation of gonadotropic function and intra-ovaric modulation of gonadal physiology
